Trastuzumab rezetecan in advanced HER2-mutant non small-cell lung cancer

The novel HER2-directed antibody-drug conjugate trastuzumab rezetecan has been shown to have a clinically meaningful activity and a good safety profile in  advanced HER2mutant non small-cell lung cancer, as reported in a new phase 2 trial recentrly published on The Lancet Oncology.

The study is a multicentr, single-arm phase 2 trial in adult patients with locally advanced or metastatic NSCLC with an activating HER2 mutation;  94 patients, who had disease progression after or were intolerant to chemotherapy and anti-PD1 or anti-PD-L1 treatment, received trastuzumab rezetecan intravenously every 3 weeks. After a median follow-up of 8.7 months, 73% of patients had a confirmed objective response; the safety profile was manageable with a low incidence of treatment discontinuation due to treatment-related
adverse events and serious events occurring in 23% of patients. As authors say, «Ttrastuzumab rezetecan at the recommended dose showed substantial and durable activity in pretreated patients with HER2-mutant NSCLC, leading to a clinically meaningful progression-free survival benefit. The activity with trastuzumab rezetecan was consistent across a range of patients with previously treated HER2-mutant NSCLC, providing a potential new treatment option for this population. The substantial and durable activity observed in this study supports further investigation of trastuzumab rezetecan in earlier disease stages».
